Product overview

What is 2-Bromo-4-Fluoro Acetanilide?

2-Bromo-4-Fluoro Acetanilide (2-B-4-FAA) is a protected halogenated aniline — an acetyl-capped 2-bromo-4-fluoroaniline. The acetyl group keeps the amine out of the way so the carbon–bromine bond can be used selectively in metalation and cross-coupling, and is removed by hydrolysis once that work is done.

With the formula C8H7BrFNO (CAS 1009-22-9), 2-B-4-FAA is a white to off-white crystalline powder melting sharply at 117–119 °C. It is practically insoluble in water but dissolves readily in methanol, dichloromethane and DMF, which are the solvents most of its downstream chemistry is run in.

It is made by acetylating 4-fluoroaniline and then brominating the resulting anilide, where the acetamido group directs the incoming bromine cleanly to the adjacent position. That regiochemistry is the whole point of the molecule: brominating the free aniline gives a much less selective result.

How it is made

4-Fluoroaniline is acetylated with acetic anhydride to 4-fluoroacetanilide, then brominated with bromine or N-bromosuccinimide under controlled temperature. The acetamido group directs substitution to the position next to it, and the product is isolated by crystallisation, washed and dried to a tight melting range.

What that means for your process

  • Sharp 117–119 °C melting range as the primary identity and purity check
  • Low isomeric bromo content and low unreacted 4-fluoroacetanilide
  • Low moisture and low residual solvent for coupling chemistry
  • Free-flowing crystalline powder for clean charging

Applications

What is 2-Bromo-4-Fluoro Acetanilide used for?

2-B-4-FAA is used as a protected building block rather than as a finished product. Its main applications, industry by industry, include:

Pharmaceutical intermediates

A selectively functionalised aniline for fluorinated heterocycle synthesis.

  • Palladium cross-coupling at the carbon–bromine bond
  • Halogen–metal exchange and quenching with electrophiles
  • Hydrolysis to 2-bromo-4-fluoroaniline when the free amine is wanted

Fluorinated heterocycles

The bromo and amino groups together close onto five- and six-membered rings.

  • Indoles and indazoles by Larock-type annulation
  • Benzimidazoles and benzothiazoles
  • Quinazolines and fused nitrogen heterocycles

Agrochemicals & fine chemicals

Fluorinated anilide motifs used across crop-protection chemistry.

  • Fluoroaniline-derived amides and ureas
  • Halogenated anilide intermediates
  • Custom protected aniline synthesis

Process route

How is 2-B-4-FAA used to build fluorinated heterocycles?

The sequence is protect, couple, deprotect, cyclise. The acetyl group arrives already in place, the bromine is consumed in a coupling or metalation step, the acetamide is then hydrolysed back to the free amine, and that amine closes the ring onto whatever was installed at the bromine position.

Deprotection

Fluorinated heterocycle

The amine cyclises onto the newly installed group to give the indole, benzimidazole or quinazoline.

Safety

Is 2-Bromo-4-Fluoro Acetanilide hazardous?

Yes. 2-B-4-FAA is harmful if swallowed, in contact with skin or if inhaled, and it irritates the eyes, skin and respiratory tract. It is a combustible solid for which no flash point has been established, so ignition sources should be controlled as a matter of course.

2-B-4-FAA should be stored in sealed drums in a cool, dry, ventilated place away from heat, light, strong oxidisers and strong bases, and handled with chemical-resistant gloves, goggles and protective clothing, with local exhaust ventilation or dust extraction at the charging point. As a fine crystalline powder it raises dust readily, so dust control is the main day-to-day exposure measure. A full MSDS is supplied with every order.

FAQ

Frequently asked questions

Common questions from buyers specifying 2-B-4-FAA for pharmaceutical, agrochemical and contract research synthesis.

What is 2-Bromo-4-Fluoro Acetanilide used for?

2-B-4-FAA is used as a protected building block for fluorinated heterocycles. The bromine is consumed in palladium cross-coupling or halogen–metal exchange, the acetyl group is then hydrolysed to free the aniline, and the resulting amine closes onto indole, indazole, benzimidazole, benzothiazole or quinazoline ring systems used in pharmaceutical and agrochemical actives.

What is the CAS number of 2-B-4-FAA?

The CAS number is 1009-22-9 and the molecular formula is C8H7BrFNO, with a molecular weight of 232.05 g/mol. Its EC number is 600-141-3 and the IUPAC name is N-(2-bromo-4-fluorophenyl)acetamide.

How is 2-Bromo-4-Fluoro Acetanilide made?

4-Fluoroaniline is acetylated with acetic anhydride to give 4-fluoroacetanilide, which is then brominated under controlled temperature. The acetamido group directs the bromine to the adjacent ring position, and the product is isolated by crystallisation.

Why protect the aniline as an acetanilide?

A free aromatic amine interferes with the chemistry that follows: it poisons palladium catalysts, is itself attacked by brominating agents, and makes regiocontrol much harder. Capping it as the acetamide tames the nitrogen, directs bromination to the position next to it, and comes off cleanly by hydrolysis once the coupling step is done.

Is 2-Bromo-4-Fluoro Acetanilide hazardous?

Yes. It is harmful if swallowed, in contact with skin or if inhaled, and irritates the eyes, skin and respiratory system. It is also a combustible solid. Handle it with PPE and dust extraction; we supply a full MSDS with every order.

How should 2-B-4-FAA be stored?

In sealed drums in a cool, dry, well-ventilated store, away from heat, direct light, strong oxidisers and strong bases. Keep containers closed between uses so the powder stays dry and free-flowing.

Do you provide COA and MSDS for 2-B-4-FAA?

Yes, we provide a batch-specific COA and MSDS with every order, plus technical support on request.

Do you supply and export 2-Bromo-4-Fluoro Acetanilide?

Yes, we supply 2-B-4-FAA across India and export internationally with complete documentation, sourced from reliable manufacturers.
